This antibody is the widely used rat anti-mouse PD-L1 clone 10F.9G2 (rat IgG2b, kappa), supplied in a low-endotoxin, RUO format intended for in vivo and functional work in mouse models. It recognizes mouse PD-L1 (CD274, UniProt Q9EP73) and is applied to flow cytometry, blocking assays, IHC on frozen sections, immunofluorescence, and functional/cell-based assays. Its established utility as a checkpoint-blocking reagent, combined with a low endotoxin burden (research grade under 1 EU/mg; ultra-low under 0.5 EU/mg) and availability in bulk milligram-to-gram quantities, makes it well suited to repeat-dose in vivo tumour immunology and T cell activation studies where endotoxin contamination would confound immune readouts. The rat IgG2b backbone is a common choice for functional PD-L1 blockade in mice, and researchers typically identify it by the 10F.9G2 clone designation. PD-L1 (programmed death-ligand 1; CD274, B7-H1) is a type I transmembrane glycoprotein of the B7 family and a principal ligand for the inhibitory receptor PD-1. Engagement of PD-1 by PD-L1 delivers a co-inhibitory signal that dampens T cell receptor signalling, curbing proliferation, cytokine production, and effector function, and contributes to peripheral tolerance and T cell exhaustion. PD-L1 is expressed constitutively on some antigen-presenting cells and is strongly upregulated on many cell types, including tumour cells and stroma, in response to inflammatory cytokines such as IFN-gamma. PD-L1 also engages CD80 (B7-1). By co-opting this pathway, tumours evade immune attack, which is why blockade of the PD-1/PD-L1 axis is a central strategy in cancer immunotherapy and a frequent target in preclinical mouse studies.
