This product is the MAR1-5A3 clone, a mouse IgG1 monoclonal antibody raised against mouse IFNAR1 (interferon-alpha/beta receptor subunit 1; UniProt P33896). MAR1-5A3 is one of the most widely used tools for blocking type I interferon signalling in mouse models, and this in-vivo formulation is prepared to be low-endotoxin (research grade <1 EU/mg; ultra-low-endotoxin <0.5 EU/mg options), supplied in bulk milligram-to-gram quantities suitable for repeated in-vivo dosing. Because it binds the ligand-binding IFNAR1 chain and neutralises signalling from multiple type I interferon subtypes at once, it is well suited to functional and in-vivo studies where the goal is to interrupt the entire type I IFN axis rather than a single interferon. Beyond in-vivo neutralisation, the clone is validated across immunoprecipitation, Western blot, flow cytometry, ELISA and blocking/functional assays, making it a flexible reagent for both mechanistic and phenotyping work. IFNAR1 is one of the two subunits of the heterodimeric type I interferon receptor, pairing with IFNAR2 to transduce signals from all type I interferons, including the many IFN-alpha subtypes and IFN-beta. IFNAR1 contributes the lower-affinity ligand-binding interactions that, together with high-affinity IFNAR2 engagement, assemble the ternary signalling complex. Receptor engagement activates the receptor-associated kinases JAK1 and TYK2, driving phosphorylation of STAT1 and STAT2, formation of the ISGF3 complex with IRF9, and transcription of interferon-stimulated genes. This pathway underlies antiviral defence, antiproliferative responses, and broad immunomodulation, and it shapes dendritic cell, T cell and NK cell function. Because type I IFN signalling is implicated in antiviral immunity, autoimmunity (notably lupus), tumour immunity and chronic inflammation, IFNAR1 is a central node for probing the biological consequences of blocking the entire type I interferon response in vivo.

Application Notes
Blocking: Clone MAR1-5A3 has a short half-life, basically because every cell expresses the IFNAR1 receptor and the receptor recycles very rapidly. And, if you want to block function in vivo, you need to be sure that all of the receptors are blocked continually in all compartments. Therefore, you need a large in vivo loading dose (2.5 mg/mouse) to saturate all the binding sites in vivo and then maintain a high enough level to keep them saturated. For in vivo blocking studies we recommend give a loading dose of 2.5 mg/mouse and follow with a weekly dose of 0.5 mg/mouse. The half-live following a 2.5 mg loading dose is about 5 days. [However, if you only inject a low dose of 250 micrograms, then the half life is 1.5 days – because you haven’t saturated the mouse]. Each investigator should determine their own optimal working dilution for specific applications.
