Background
This is clone HDB-4A7, a mouse monoclonal antibody of the IgG2a isotype raised against mouse interferon beta (IFN beta). Because the immunogen, host, and target are all murine, the antibody is well suited to in-vivo neutralization studies in mouse models without the confounding anti-species immune responses that can accompany rat or hamster reagents used long term. HDB-4A7 recognizes and functionally blocks bioactive mouse IFN beta, and is validated for Western blot, blocking, and functional (neutralization) assays, making it useful both for detecting the cytokine and for interrogating its biological role in vivo. The antibody is supplied low-endotoxin (research grade below 1 EU/mg, ultra-low-endotoxin option below 0.5 EU/mg) and in bulk milligram-to-gram quantities, so that repeated dosing across treatment cohorts is feasible while minimizing endotoxin-driven artefacts that would otherwise confound type I interferon readouts. It is provided for research use only (RUO) and is intended as a tool for controlled laboratory investigation rather than any diagnostic or therapeutic application. Interferon beta (IFN beta, gene Ifnb1; UniProt P01575 in mouse) is a type I interferon and a central cytokine of innate immunity. It is induced rapidly upon sensing of viral and microbial nucleic acids and other danger signals through pattern-recognition pathways, and unlike the multiple IFN alpha subtypes it is encoded by a single gene. Secreted IFN beta signals in an autocrine and paracrine manner by binding the shared type I interferon receptor (IFNAR1/IFNAR2), triggering JAK-STAT signaling and transcription of interferon-stimulated genes that establish an antiviral state. Beyond antiviral defense, IFN beta shapes immune responses by influencing dendritic cell maturation, T cell and NK cell activity, and antitumor immunity, and it has context-dependent roles in autoimmunity and chronic inflammation. Because type I interferon signaling can be either protective or pathogenic depending on setting, neutralizing IFN beta specifically is a valuable way to dissect its distinct contribution.

Blocking: Blocks in vivo IFNb leading to increased the lethality of mice infected with West Nile Virus (WNV), similar to the susceptibility of IFNb-/- mice. Clone HDB-4A7 circulates with a half-life of ~ 14 days. Each investigator should determine their own optimal working dilution for specific applications.
