Background
This is the IM7 clone, a rat IgG2b monoclonal raised against mouse CD44. IM7 is one of the most widely used anti-CD44 antibodies and reacts with an epitope shared by all known mouse CD44 isoforms (the standard form and CD44 variant splice forms), making it a broad pan-CD44 reagent. The rat IgG2b format is well suited to in-vivo and functional work in mice, where cross-reactivity and Fc handling are important considerations. This preparation is offered as a low-endotoxin, in-vivo-ready formulation (research grade <1 EU/mg; ultra-low <0.5 EU/mg) and is available in bulk milligram-to-gram quantities to support repeat dosing and multi-cohort studies. It is validated across a range of applications including flow cytometry, CyTOF, immunoprecipitation, Western blot, frozen-section IHC, ELISA, and functional assays, and is commonly used to track and manipulate CD44-expressing populations such as activated and memory T cells. CD44 is a widely expressed single-pass transmembrane glycoprotein and the principal cell-surface receptor for hyaluronan, though it also binds osteopontin, collagens, matrix metalloproteinases, and other extracellular matrix components. Extensive alternative splicing of variant exons and heavy post-translational glycosylation generate a large family of isoforms, from the ubiquitous standard form (CD44s) to tissue- and activation-restricted variant forms (CD44v). Through hyaluronan engagement and links to the cytoskeleton, CD44 contributes to cell adhesion, migration, and signaling. In immunology it is a classic marker of lymphocyte activation and memory: naive T cells are CD44-low, whereas antigen-experienced effector and memory T cells upregulate CD44, and CD44 participates in leukocyte rolling, extravasation, and homing. CD44 is also implicated in tumor invasion, metastasis, and cancer stem-cell biology, which is why it is studied both as a phenotypic marker and as a functional target.
