ichorRed® Nucleic Acid Gel Stain (10,000x in water)

Background:

ichorRed® Nucleic Acid Gel Stain is the third generation non-mutagenic fluorescent nucleic acid gel stain to replace the highly toxic traditional ethidium bromide (EB) or the expensive GelRed® (GR) from Biotium for staining dsDNA, ssDNA or RNA in agarose gels or polyacrylamide gels.
ichorRed® is more sensitive than EB and GR either as precast gel stains or post gel stains.

We can directly replace EB or GR with ichorRed® without changing the existing imaging system since they all have the similar spectra. ichorRed® can also be used to stain dsDNA, ssDNA or RNA in polyacrylamide gel via post gel staining. Gel staining with ichorRed® is compatible with downstream applications such as gel extraction and cloning. ichorRed® is efficiently removed from DNA by phenol/chloroform extraction and ethanol precipitation.

Precast Agarose Gel Protocol:
Prepare molten agarose gel solution using your standard protocol.
Dilute ichorRed® 10,000X stock reagent into the molten agarose gel solution at 1:10,000 (e.g. 5ul added to 50ml of the gel solution) and mix
thoroughly. ichorRed® can be added while the gel solution is still hot.
Cast the gel and allow it to solidify. Any leftover gel solution may be stored and reheated later for additional gel casting.
ichorRed® precast gels may be stored at 4°C for later use.
Load samples and run the gels using your standard protocol.
ichorRed®: Image the stained gel with a standard transilluminator (302 or 312nm) use UV light box with EB filter.
Post-staining agarose or acrylamide gels:

Dilute the ichorRed® 10,000X stock reagent~3,300 fold to make a 3X staining solution in H2O. Note: including 0.1M NaCl in the staining solution enhances sensitivity, but may promote dye precipitation if the gel stain is reused.Carefully place the gel in a suitable polypropylene container. Gently add a sufficient amount of the 3X staining solution to submerge the gel.Agitate the gel gently at room temperature for 30 minutes.

ichorRed®: Image the stained gel with a standard transilluminator (302 or 312nm), and photograph the gel using an ethidium bromide filter

Staining solution can be resued 2-3 times. Store staining solution at room temperature protected from light.

AMES Test:
ichorRed® was negative in the Ames Test using Salmonella strain TA97, TA98, TA100 and TA102 (with/without S9 metabolic activation), as the revertant colonies number is less than two times of the initiative revertant colonies number with/without S9 metabolic activation. The same data was obtained by the repeated experiment
Aquatic Toxicity Test:
Both ichorRed® at 3x concentration is classified as nonhazardous to aquatic life, under CCR Title 22 regulation. ichorRed® and ichorGreen at 3x or lower concentrations can be safely released into the environment.
