Clone B8-24-3 is a mouse-derived monoclonal antibody (mouse IgG1, kappa) that recognizes the mouse MHC class I H-2b haplotype alloantigen. The hybridoma (ATCC TIB-139) was raised against H-2b-bearing cells; the antibody binds cells of the H-2 b haplotype and cross-reacts with H-2p/H-2pa, while remaining negative on d, f, j, k, q, s, v, u and w haplotypes, giving it clean haplotype-level discrimination. Because it targets a cell-surface alloantigen rather than an intracellular protein, B8-24-3 is well suited to in-vivo and functional immunology work: allorecognition and transplantation models, studies of antigen presentation to CD8+ T cells, and identification or manipulation of H-2b-positive populations in chimeras and adoptive-transfer settings. H-2b is the classical MHC class I haplotype of C57BL/6 and related mouse strains, encoded at the H-2K, H-2D and H-2L loci. MHC class I molecules are heterodimers of a polymorphic heavy chain non-covalently associated with beta-2-microglobulin. They are expressed on nearly all nucleated cells and continuously present short (8-10mer) peptides derived from intracellular proteins to CD8+ cytotoxic T cells via the T-cell receptor. This surveillance pathway lets the immune system detect virally infected, transformed, or otherwise abnormal cells, and MHC class I alloantigens are central to allorecognition and graft rejection. The polymorphic residues that define the b haplotype (H-2Kb, H-2Db) shape both peptide-binding specificity and allogeneic recognition, making haplotype-specific antibodies useful tools for identifying, tracking, and functionally probing H-2b-bearing cells.
