This bulk anti-human CD25 antibody is the mouse monoclonal clone 7G7B6, a mouse IgG2a that recognizes the human interleukin-2 receptor alpha chain (IL-2Rα/CD25, UniProt P01589). 7G7B6 is a long-established, widely characterized clone binding an epitope on human CD25 that is distinct from the IL-2 binding site, making it useful for detecting receptor expression without necessarily competing with ligand. It is validated across a broad application set (ELISA, flow cytometry, immunofluorescence, immunoprecipitation, and Western blot), supporting both surface phenotyping and biochemical characterization of the receptor. CD25 is the 55 kDa alpha chain of the interleukin-2 receptor (IL-2Rα, Tac antigen; UniProt P01589). On its own CD25 binds IL-2 with low affinity; when it assembles with the IL-2Rβ (CD122) and common gamma (CD132) chains it forms the high-affinity heterotrimeric receptor that drives IL-2 signaling through JAK/STAT5, PI3K, and MAPK pathways. CD25 is constitutively high on CD4+FOXP3+ regulatory T cells, where it is a defining marker and supports Treg fitness and immune homeostasis, and it is rapidly upregulated on conventional T and B lymphocytes following activation, making it a classic activation marker. CD25 is also expressed on some leukemias and lymphomas. A soluble form is shed into serum and can serve as an immune-activation biomarker. Because of its central role in T cell tolerance and expansion, CD25 is a well-studied target in autoimmunity, transplantation, and oncology.
