Background
This product is a purified rat anti-mouse Integrin beta 7 (ITGB7) monoclonal antibody, clone FIB504, of the rat IgG2a kappa isotype. FIB504 is a widely used, well-characterized clone that recognizes the mouse beta 7 integrin subunit and blocks its adhesive function, making it suited to in-vivo and functional studies of lymphocyte trafficking and gut/mucosal homing rather than purely descriptive work. The antibody is validated for flow cytometry and for functional blocking applications. It is manufactured to a low-endotoxin specification (research grade <1 EU/mg; ultra-low <0.5 EU/mg) and supplied in bulk milligram-to-gram quantities, which supports repeated dosing across treatment groups and longer in-vivo studies where endotoxin contamination would otherwise confound immune readouts. As a rat IgG2a, it pairs naturally with a matched rat IgG2a isotype control for in-vivo experiments. All material is for research use only (RUO). Researchers typically identify this reagent by its clone designation, FIB504, when comparing lots and vendors. Integrin beta 7 (ITGB7, UniProt P26011) is an integrin beta subunit that pairs with alpha chains to form two heterodimers central to mucosal immunity. With alpha 4 (ITGA4) it forms alpha4beta7 (LPAM-1), the gut-homing integrin that binds MAdCAM-1 on intestinal and gut-associated lymphoid endothelium, directing lymphocyte trafficking to the gut and Peyer's patches. With alpha E (CD103, ITGAE) it forms alphaEbeta7, which binds E-cadherin on epithelial cells and retains intraepithelial lymphocytes and tissue-resident memory T cells within mucosal epithelium. Because beta 7 is shared by both heterodimers, it is a key determinant of intestinal immune homeostasis, oral tolerance, and mucosal inflammation. The alpha4beta7/MAdCAM-1 axis is a validated therapeutic target in inflammatory bowel disease, and beta 7 also contributes to lymphocyte retention in tissues such as the intestinal lamina propria.
