Background
Anti-Mouse Tim-3 clone RMT3-23 is a rat IgG2a kappa monoclonal antibody that recognises mouse Tim-3 (HAVCR2; UniProt Q8VIM0). Raised in rat against the mouse antigen, it is a widely used research reagent for detecting and functionally interrogating Tim-3 on exhausted and activated T cells, regulatory T cells, and myeloid populations. The clone is validated for flow cytometry, Western blot, blocking, and IHC on frozen sections, making it suitable both for characterising Tim-3 expression and for functional in-vivo and ex-vivo studies of Tim-3 biology. ichor.bio supplies RMT3-23 in a low-endotoxin, in-vivo-ready format (research grade <1 EU/mg; ultra-low <0.5 EU/mg), so it is appropriate for antibody-administration experiments in mice where endotoxin can confound immune readouts. It is produced at scale (bulk milligram to gram quantities) to support dosing across cohorts and longitudinal studies. Because researchers most often search by clone, RMT3-23 is the key identifier for this product. All material is for research use only (RUO) and is not intended for diagnostic or therapeutic use. Tim-3 (T-cell immunoglobulin and mucin-domain containing-3), encoded by Havcr2, is a type I membrane protein and immune checkpoint receptor. It is expressed on exhausted and terminally differentiated CD8+ and CD4+ T cells, regulatory T cells, NK cells, and cells of the myeloid lineage including dendritic cells and macrophages. Tim-3 has an IgV domain that engages several ligands, including galectin-9, phosphatidylserine, HMGB1, and CEACAM1, and signalling generally delivers inhibitory, T-cell-dampening cues that limit effector function. On chronically stimulated T cells in tumours and persistent viral infection, Tim-3 is frequently co-expressed with PD-1 and marks a deeply exhausted state. Because of this role in restraining anti-tumour and antiviral immunity, Tim-3 is an actively studied immune-checkpoint target, and antibodies against it are used to probe checkpoint blockade, T-cell exhaustion, and combination immunotherapy in preclinical models.
