This is a purified rat monoclonal antibody, clone TY25 (rat IgG2a), that recognises mouse PD-L2 (Programmed Death-Ligand 2, also called PD-L2/B7-DC/CD273; UniProt Q9WUL5). It is validated for blocking as well as Western blot, flow cytometry, frozen IHC and immunoprecipitation. The rat IgG2a isotype and the neutralising, receptor-blocking activity of TY25 make it well suited to in-vivo and functional studies in mouse models where the goal is to interrupt PD-L2 engagement of its receptors and dissect the relative contribution of PD-L2 versus PD-L1 to PD-1-mediated tolerance. The preparation is supplied at low endotoxin (research-grade <1 EU/mg; ultra-low-endotoxin <0.5 EU/mg options) and in bulk milligram-to-gram quantities, formats intended for repeat-dosing tumour, infection, autoimmunity and airway/allergy models rather than single-well assays. PD-L2 (B7-DC/CD273) is one of the two ligands for the inhibitory receptor PD-1, the other being PD-L1. Compared with the broadly expressed PD-L1, PD-L2 has a more restricted expression pattern, found chiefly on dendritic cells, macrophages, and some B cells, and it is strongly induced by Th2 cytokines such as IL-4. PD-L2 binds PD-1 with higher affinity than PD-L1 does, and engagement delivers inhibitory signals that attenuate T-cell receptor signalling, cytokine production and proliferation, contributing to peripheral tolerance and to the resolution of immune responses. PD-L2 also interacts with RGMb (repulsive guidance molecule b), a second receptor implicated in respiratory tolerance. In tumours and in chronic infection PD-L2 can restrain effector T-cell responses, making it a distinct axis of the PD-1 checkpoint that is often studied alongside, or in contrast to, PD-L1.
