This antibody targets mouse IL-4 using clone 11B11, a rat IgG1 kappa monoclonal raised against the murine cytokine. 11B11 is one of the most widely used and long-established anti-mouse IL-4 clones, valued for its ability to neutralize IL-4 bioactivity and for its versatility across capture ELISA, ELISPOT, flow cytometry (including intracellular staining), CyTOF, immunoprecipitation, IHC, and functional/neutralization assays. The rat IgG1 backbone makes it suitable for in-vivo cytokine-blockade studies in mice, where a non-depleting, neutralizing format is preferred. This preparation is manufactured for in-vivo and functional research: it is low endotoxin (research grade <1 EU/mg; ultra-low <0.5 EU/mg), supplied in bulk milligram-to-gram quantities, and provided without preservatives or carrier proteins that could confound animal work. It is for research use only (RUO). These attributes suit studies of Th2 immunity, allergy and asthma models, helminth infection, tissue repair, and macrophage polarization, where reproducible, endotoxin-controlled reagent is important for interpreting immune readouts. IL-4 is a pleiotropic type I cytokine and the signature effector of type 2 (Th2) immunity. It is produced chiefly by activated CD4+ Th2 cells, but also by basophils, mast cells, eosinophils, and group 2 innate lymphoid cells (ILC2s). IL-4 signals through the type I receptor (IL-4Ralpha paired with the common gamma chain) on hematopoietic cells and the type II receptor (IL-4Ralpha with IL-13Ralpha1) on non-hematopoietic cells, activating JAK/STAT6 signaling. Its major functions include driving naive CD4+ T-cell differentiation toward the Th2 lineage, promoting B-cell immunoglobulin class switching to IgE and IgG1, and polarizing macrophages toward an alternatively activated (M2) phenotype. IL-4 is central to allergic inflammation, asthma, and host defense against helminths, and contributes to tissue remodeling and fibrosis. Because IL-4 and IL-13 share IL-4Ralpha, their activities partially overlap, though IL-4 dominates early Th2 commitment.

Application Notes
ELISA Sandwich: This antibody is useful as the capture antibody in a sandwich ELISA. The suggested coating concentration is 0.5-2.0 µg/ml. ELISPOT Capture: This Low Endotoxin Functional Formulation antibody has been reported for use as the capture antibody in a sandwich ELISPOT assay. Aseptically coat plates at a concentration of 2-6 µg/ml is recommended. Neutralization: Anti-Mouse IL-4 (clone 11B11) antibody can neutralize the bioactivity of natural or recombinant IL-4. The Low Endotoxin Functional Formulation purified antibody (Endotoxin <0.1 EU/μg, Azide-Free, 0.2 μm filtered) is recommended for neutralization of mouse IL-4 bioactivity in vivo. Each investigator should determine their own optimal working dilution for specific applications.
