Clone B122 is an Armenian hamster IgG monoclonal antibody raised against mouse interleukin-1 beta (IL-1b). Armenian hamster antibodies against mouse cytokines are widely used for in-vivo work because their framework is sufficiently divergent from mouse to generate a specific anti-cytokine response yet is generally well tolerated across repeated dosing in mouse models. B122 recognizes mouse IL-1b and neutralizes its bioactivity, and it is also validated for immunoprecipitation, western blot, and ELISA, making it suitable both for functional cytokine-blockade studies and for detecting and quantifying IL-1b in biological samples. This preparation is supplied at low endotoxin (research-grade <1 EU/mg; ultra-low <0.5 EU/mg) and in bulk milligram-to-gram quantities, which is important for in-vivo studies where endotoxin contamination would itself trigger IL-1 pathway activation and confound results. It is offered for research use only (RUO). Researchers typically select B122 when they need to interrogate the contribution of IL-1b signaling to inflammation, host defense, autoimmunity, or tumor biology in mouse models without the confounding effects of endotoxin. Interleukin-1 beta (IL-1b, gene Il1b; UniProt P10749) is a central pro-inflammatory cytokine of the IL-1 family. It is synthesized as an inactive precursor (pro-IL-1b) that lacks a signal peptide and is proteolytically matured, classically by caspase-1 following inflammasome (e.g., NLRP3) activation, then released via non-classical secretion. Mature IL-1b signals through the type I IL-1 receptor (IL-1R1) together with the accessory protein IL-1RAcP, driving MyD88-dependent NF-kB and MAPK signaling. Its activity is naturally restrained by IL-1 receptor antagonist (IL-1Ra) and the decoy receptor IL-1R2. IL-1b induces fever, endothelial activation, leukocyte recruitment, and downstream cytokines, and contributes to Th17 responses. Dysregulated IL-1b drives autoinflammatory disease, and it is implicated in autoimmunity, metabolic inflammation, and tumor-promoting inflammation, making it a heavily studied therapeutic and mechanistic target.
