Background
This product is clone ALF-161, an Armenian Hamster IgG1 monoclonal antibody raised against mouse IL-1a (interleukin-1 alpha, UniProt P01582). As a hamster anti-mouse reagent, it is well suited to in-vivo neutralization studies and functional work in mouse models, where it binds IL-1a and blocks its interaction with the IL-1 receptor. ALF-161 is widely used for in-vivo IL-1a neutralization and across in-vitro applications including Western blot, ELISA, ELISPOT, immunoprecipitation, and intracellular flow cytometry. This in-vivo formulation is manufactured to low endotoxin levels (research grade typically less than 1 EU/mg; ultra-low options less than 0.5 EU/mg) to minimize confounding innate immune activation during systemic administration. It is supplied in bulk quantities (milligram to gram scale) suitable for repeated-dosing regimens and larger animal cohorts. All material is for research use only (RUO). Researchers commonly identify this reagent by its clone designation, ALF-161, when comparing across suppliers. IL-1a (interleukin-1 alpha) is a pro-inflammatory cytokine of the IL-1 family and, with IL-1b, is one of the two principal IL-1 receptor agonists. Unlike IL-1b, IL-1a is constitutively expressed by many cell types (epithelial cells, keratinocytes, fibroblasts, endothelial cells) and is present as a precursor that is biologically active without caspase-1 processing. It localizes to the cytosol and nucleus and can be released upon cell death or necrosis, acting as an alarmin (DAMP) that signals tissue damage. Both membrane-associated and secreted forms engage the type I IL-1 receptor (IL-1R1) together with the co-receptor IL-1RAcP, driving MyD88/NF-kB signaling and downstream inflammation. IL-1a contributes to sterile inflammation, fever, leukocyte recruitment, and tumor-associated inflammation, making it a target of interest in inflammatory disease and cancer models.
