Background
This product is JAMA-147, an Armenian hamster IgG monoclonal antibody raised against mouse IL-1R1 (interleukin-1 receptor type 1, UniProt P13504). Because it is a hamster anti-mouse reagent that binds the receptor directly, it is well suited to functional and in-vivo studies in mouse models where blocking IL-1 signalling is the goal, and it also supports immunoprecipitation, western blot, and flow cytometry for target detection and validation. Hamster IgG framework antibodies are widely used in mouse work because they are recognised as foreign only weakly, allowing repeat dosing while limiting confounding host immune responses. The preparation is manufactured to low-endotoxin specifications (research grade under 1 EU/mg; ultra-low grade under 0.5 EU/mg), which is important for in-vivo experiments where contaminating endotoxin would itself trigger innate immune and IL-1-family responses and confound results. IL-1R1 (IL1R1, CD121a; UniProt P13504) is the primary signalling receptor for the interleukin-1 family cytokines IL-1 alpha and IL-1 beta. It is a type I transmembrane glycoprotein with three extracellular immunoglobulin-like domains and a cytoplasmic Toll/IL-1 receptor (TIR) domain. Productive signalling requires ligand-induced recruitment of the accessory protein IL-1RAcP (IL1RAP), which assembles a ternary complex that engages the adaptor MyD88, IRAK kinases, and TRAF6, driving activation of NF-kappaB and MAPK pathways and downstream pro-inflammatory gene expression. Signalling is naturally restrained by the secreted decoy receptor IL-1R2 and by the endogenous antagonist IL-1Ra. IL-1R1 is broadly expressed, including on fibroblasts, endothelial and epithelial cells, and subsets of immune cells, and its activity is central to sterile and infectious inflammation, fever, and autoinflammatory disease. It is therefore a frequent target in inflammation, autoimmunity, and tumour microenvironment research.
