Background
This is clone Y-3, an antibody directed against mouse H-2K, a classical MHC class I molecule. The Y-3 clone was originally raised by immunising a BALB/c (H-2d) mouse with concanavalin-A-stimulated spleen cells from the congenic BALB.B (H-2b) strain, and it reacts with H-2K across several haplotypes (b, k, q, r, s) while not binding H-2d. The clone is well established in immunology as a tool for identifying, blocking and immunoprecipitating H-2K class I heavy chains. ichorbio supplies it in an in-vivo-ready format: purified, low endotoxin (<1 EU/mg research grade, with an ultra-low <0.5 EU/mg option), and available in bulk milligram-to-gram quantities suited to repeat-dosing mouse studies. Validated applications include Western blot and functional/blocking assays. These attributes make it appropriate for functional MHC class I work in vivo and in vitro, such as blocking CD8 T-cell recognition, transplantation and allorecognition models, and cell-surface class I detection. For research use only; not for diagnostic or therapeutic use. H-2K is one of the two classical MHC class I loci in the mouse (alongside H-2D), the murine counterpart of human HLA class I. It encodes a polymorphic heavy chain that pairs non-covalently with beta-2 microglobulin and presents short (typically 8-10 residue) endogenously derived peptides at the cell surface. This peptide-MHC complex is surveyed by the T-cell receptor of CD8 cytotoxic T lymphocytes, allowing the immune system to detect intracellular pathogens and tumour-associated antigens. H-2K is highly polymorphic between mouse strains, defining haplotypes (e.g. H-2Kb in C57BL/6), which underlies its central role in allorecognition and transplant rejection. Class I molecules also serve as ligands for inhibitory and activating NK-cell receptors, contributing to "missing-self" recognition. Because it is expressed on nearly all nucleated cells, H-2K is a widely used marker of MHC class I biology and antigen presentation.
