Background
This is a bulk-format mouse monoclonal antibody (clone UCHT-4, isotype Mouse IgG2a) raised against human CD8, the alpha chain of the CD8 co-receptor (UniProt P01732). Because it is a well-characterised anti-human CD8 clone, it is suited to identifying, quantifying, sorting and functionally probing human cytotoxic and CD8-positive T cells across a broad set of applications, including flow cytometry, Western blot, immunoprecipitation, IHC, immunofluorescence, ELISA and functional assays. Supplied as a research-use-only (RUO) reagent in bulk quantities (mg to gram scale), it is intended for laboratories that consume large amounts of a single validated lot and want lot-to-lot consistency for screening, assay development or reagent manufacturing workflows. Low-endotoxin formulation makes it appropriate for sensitive primary-cell and functional work where endotoxin contamination could confound results. As a human-reactive antibody, it is used primarily in vitro and ex vivo on human samples rather than in mouse in-vivo dosing studies. CD8 is a cell-surface glycoprotein co-receptor central to the cytotoxic arm of adaptive immunity. On conventional cytotoxic T lymphocytes it exists as an alpha/beta heterodimer, while CD8 alpha/alpha homodimers appear on subsets of NK cells, dendritic cells and intraepithelial lymphocytes. The CD8 co-receptor binds the largely invariant alpha-3 domain of MHC class I molecules, stabilising the interaction between the T-cell receptor and peptide-MHC class I on target cells. Through its cytoplasmic tail CD8 alpha recruits the Src-family kinase LCK, bringing this kinase into proximity with the TCR-CD3 complex to initiate proximal signalling and lower the threshold for T-cell activation. CD8 expression thus marks and enables the T cells responsible for recognising and killing virally infected and tumour cells, making it one of the most widely used lineage markers in human immunology and immuno-oncology.

Specificity
Anti-CD8 In Vivo Antibody - Low Endotoxin (UCHT-4) recognizes Human CD8. Anti-Human CD8 recognizes a T-cell glycoprotein cell surface complex (Mr 32 kDa) which defines the cytotoxic/suppressor population of mature T-lymphocytes. The CD8 complex interacts with class I MHC molecules resulting in increased adhesion between CD8+ T-lymphocytes and their target cells. Binding of the CD8 antigen to Class I MHC molecules enhances T-cell activation. The cytoplasmic portion of the CD8 antigen is associated with a protein tyrosine kinase (p56lck).
