Anti-Mouse CD120a clone 55R-170 is an Armenian hamster IgG1 monoclonal raised against mouse CD120a (TNF receptor 1, TNFR1/p55). Armenian hamster IgG mAbs are widely used for in vivo work against mouse targets because they are well tolerated over repeated dosing and their non-mouse origin gives clean, low-background staining in mouse systems. This preparation is validated across immunoprecipitation, Western blot, flow cytometry, and functional/blocking and neutralization assays, making it suitable for both mechanistic characterization and receptor-level intervention studies. It is manufactured to a low-endotoxin research grade (<1 EU/mg; ultra-low <0.5 EU/mg) and supplied in bulk milligram-to-gram quantities, so that endotoxin-driven artefacts are minimized in sensitive in vivo TNF-signaling models where background inflammation must be controlled. The antibody is aggregate-screened and buffered without carrier proteins or preservatives that interfere with injection. It is provided for research use only (RUO). These attributes make clone 55R-170 appropriate for investigators dissecting TNFR1-dependent inflammation, cell death, and immune signaling in mouse models, and for repeated-dose in vivo receptor-blocking studies. CD120a, better known as tumor necrosis factor receptor 1 (TNFR1; also p55/p60), is encoded by Tnfrsf1a and is the principal receptor mediating TNF-alpha signaling. It is expressed broadly on most nucleated cell types, in contrast to the more restricted TNFR2 (CD120b). TNFR1 binds both soluble and membrane TNF and, uniquely among the two receptors, contains a cytoplasmic death domain. Ligand engagement triggers assembly of complex I (TRADD, TRAF2, RIPK1, cIAPs) that activates NF-kB and MAPK pathways to drive pro-inflammatory and pro-survival gene expression; under conditions of altered ubiquitination or checkpoint loss, signaling shifts to complex II, promoting caspase-8-dependent apoptosis or RIPK3/MLKL-driven necroptosis. Through these outputs TNFR1 governs inflammation, host defense, tissue homeostasis, and cell-fate decisions, and is central to models of septic shock, autoimmune and inflammatory disease, and inflammation-driven pathology.
